Screening and selection of antibody producing clones is done using a special medium called HAT medium. In this medium only hybridoma cells that are a formed by fusion of antibody producing B-lymphocytes and Myeloma cells and can survive the nucleic acid synthesis blocking agent aminopterin in the HAT medium. Normal cells can utilize the salvage pathway to produce nucliec acids, but are not immortal unlike myeloma cell lines.
